PROTAC

PROTAC (Proteolysis Targeting Chimera) is a bifunctional small molecule degrader composed of three key elements: a ligand that binds to the target protein, a ligand that recruits an E3 ubiquitin ligase, and a linker connecting the two. Unlike traditional inhibitors that merely block protein activity, PROTAC molecules facilitate the ubiquitination and degradation of target proteins via the ubiquitin-proteasome pathway. This technology enables efficient targeting of traditionally “undruggable” proteins and overcomes drug resistance mechanisms, offering broad applications in cancer, neurodegenerative diseases, and rare disorders with advantages such as high catalytic efficiency, sustained effects, and a modular design platform.
